Irigenin exhibits anticancer activity against human colon cancer cells via autophagy, inhibition of cell migration and invasion, and targeting of ERK/MAPK signal pathway

 Purpose: To study the anticancer effect of naturally-occurring irigenin isoflavonoid on colon cancer, and to determine mechanism involved.
 Methods: The viability normal cancerous cells was assessed by MTT assay, while clonogenic assay used measure colony generation. Autophagy examined transmission electron microscopy (TEM) western blotting. Transwell chamber influence cell migration invasion. expression levels ERK/MAPK signal pathway-associated proteins were assayed using Western blotting.
 Results: Irigenin significantly decreased Caco-2 cancer cells, in contrast CCD841 produced concentration-dependent anti-proliferative effects (p < 0.05). number colonies control group 0.05) upon exposure irigenin. Results from TEM revealed that caused dose-dependent formation autophagosomes, dose-based up-regulation expressions Beclin-1, LC3-I LC3-II Moreover, markedly suppressed invasion Coca-2 cells.  Conclusion: These results indicate exerts potent inhibitory growth induces autophagy, inhibits invasion, targets survival pathway. Therefore, may be a lead candidate drug for treatment. However, there is need further vivo clinical studies validate these findings.
